I'm definitely technology challenged enough that if I were to even attemp these, they would probably wind up being irreversible. Happy to say I am not a nerd.
My favorite--use Keynote (Mac) or PowerPoint to create a slide that's an exact duplicate of a real work session--a Word document, maybe part of a email behind it, etc.
Program the slide with a scary transition like a poof of light, and then go to an all black slide.
Leave it on their desktop in full screen view. When they come back and click, their whole desktop will look like it just exploded.
Variation: set a timer on the second (black) slide to go to "April Fools" after 10-20 seconds.
i have to say this is one day that I don't really like being a teacher. I have kids coming up to me saying my hair is messed up or I have a big booger on my nose or that some other teacher needs to talk to me. I threatened them with detention, maybe I should tell them, I'm only kidding too :-)
The teachers are just as bad! The English teacher for our homeschool co-op sent out an e-mail today telling the kids they had all missed a deadline yesterday and also that they all needed to write 35 compound-complex sentenced by this afternoon. There was SERIOUS weeping and gnashing of teeth until the April Fools' e-mail came through!
I should have NEVER sent this "Pranks for Nerds" link to my techie son! He will not get anything else done today :( (I'm sure my husband is now not getting anything done at work either!)
I flipped the screen on my wife's account to the right and let her try to navigate for about five minutes. I'm glad she's a patient woman. Otherwise, my life might have been in danger.

Here's another fun prank. On some PCs, pressing CTRL+ALT+UPARROW (or CTRL+ALT+DOWNARROW) will flip the display upside down.
